چکیده
Immune cells have a remarkable influence on tissue homeostasis and stem cell function. Our study is focused how age-related immunometabolic alterations in T can affect fundamental dependent processes the skin, such as skin homeostasis, hair growth regeneration. To address this, we used Tfamfl/fl Cd4Cremouse model with severe mitochondrial dysfunction owing to lack of transcription factor A (Tfam). In absence Tfam, which nuclear-encoded protein essential for DNA stabilization replication, metabolism impaired this are forced undergo shift towards glycolysis. This leads aberrant cell-driven cytokine cytotoxic responses. results indicated that these mice show signs premature aging well altered abundance bulge cells, altogether suggesting defective mobilization. Furthermore, functional experiments revealed hampered regrowth wound healing. Further using bone marrow transplantation supported faulty were responsible Mechanistically, large-scale molecular analyses dramatic changes epigenetic landscape transcriptomic profile consequence sustained glycolytic cells. Overall, our point at deficient follicle function ultimately caused by metabolism, turn promotes appearance signs.

Aging in hair follicle stem cells and niche microenvironment.
Hair graying and hair loss are prominent and common characteristics of the elderly population. In some individuals these processes can significantly impact their quality of life, leading to depression, anxiety and other serious mental health problems. Accordingly, there has been much interest in understanding the complex physiological changes within the hair follicle in the aging individual. It...
متن کاملMitochondrial aerobic respiration is activated during hair follicle stem cell differentiation, and its dysfunction retards hair regeneration
Background. Emerging research revealed the essential role of mitochondria in regulating stem/progenitor cell differentiation of neural progenitor cells, mesenchymal stem cells and other stem cells through reactive oxygen species (ROS), Notch or other signaling pathway. Inhibition of mitochondrial protein synthesis results in hair loss upon injury. However, alteration of mitochondrial morphology...

Background: Hair follicle stem cells (HFSCs) located in the bulge area has shown to be highly proliferative and could differentiate into neurons, glia, smooth muscle cell, and melanocytes in vitro. Simvastatin is an HMG-CoA reductase inhibitor that exerts pleiotropic effects beyond simple low-density lipoprotein lowering and has a similar impact on the differentiation of bone marrow stromal cel...
متن کاملEstablishment of human hair follicle mesenchymal stem cells with overexpressed human hepatocyte growth factor
Objective(s): Chronic liver disease has become a major health problem that causes serious damage to human health. Since the existing treatment effect was not ideal, we need to seek new treatment methods. Materials and Methods: We utilized the gene recombination technology to obtain the human hair mesenchymal stem cells which overexpression of human hepatocyte growth factor (hHGF). Furthermore, ...
